    The effect of problem type on the strategies used by novice designers

    Previous research by the author demonstrated that untutored Year 7 students produce a solution to a design-and-make task in ways significantly different to those prescribed by textbooks. However, the strategy used may have been a function of the particular design brief and how it was presented. The follow-up study described here addressed the question: Is the design strategy used by novices dependent on the task? Four Year 7 dyads were video recorded while designing and making a solution to a specific task. Comparisons were made between dyads in the current study, between each dyad and a map of the theoretical model, and between dyads in both studies. Analysis showed no significant difference between dyads in the current study, nor between dyads in the two studies, but confirmed significant differences between dyads' strategies and those described in the literature. Students did not use two-dimensional modelling to explore and communicate a design proposal, nor did they generate multiple solutions. They moved immediately to three-dimensional modelling, but often lacked the requisite skills to develop their ideas.     Teaching sketching and its effect on the solutions produced by novice designers

    Previous research by the authors showed that novice designers do not use sketching as a way to explore and communicate a design proposal, but move immediately to three-dimensional modelling. Neither do they generate multiple solutions. Furthermore, they lack the skills to readily explore their ideas and communicate them to others. The follow-up study described here addressed the questions: How can two-dimensional modelling be taught to students so they are better able to express their ideas? What is the relationship between the ability of a student to make two-dimensional models and the quality of design proposals? What specific skills, knowledge and materials are required for both teaching and learning sketching? Sixteen Year 7 students were divided into eight single-sex dyads. Four received instruction in freehand sketching. Four received no instruction and acted as a control group. The eight dyads were videotaped while producing a solution to a common design brief. Analysis of the data has provided insights into the effects of instruction on the proposals produced by students, as well as feedback on the efficacy of a set of materials for teaching sketching

    Developing a standardized measure of short-term memory and syntactic complexity: results from subtests of the CRTT-R

    Short-term memory (STM) effects have shown to be distinguishable from other working memory components supporting complex computations/central executive functions. To develop a measure capable of assessing the effects of STM and linguistic computations on sentence processing,  effects of syntactic complexity and padding were investigated with the Computerized Revised Token Test –Revised in individuals with aphasia and control participants.  Off-line measures revealed clear effects of both factors.  The expected interaction of complexity and padding  and overadditive effects for individuals with aphasia were not found. An effect of complexity on word errors in passive sentences for individuals was shown

    A simple device for multiplex ELISA made from melt-extruded plastic microcapillary film

    We present a simple device for multiplex quantitative enzyme-linked immunosorbant assays (ELISA) made from a novel melt-extruded microcapillary film (MCF) containing a parallel array of 200µm capillaries along its length. To make ELISA devices different protein antigens or antibodies were immobilised inside individual microcapillaries within long reels of MCF extruded from fluorinated ethylene propylene (FEP). Short pieces of coated film were cut and interfaced with a pipette, allowing sequential uptake of samples and detection solutions into all capillaries from a reagent well. As well as being simple to produce, these FEP MCF devices have excellent light transmittance allowing direct optical interrogation of the capillaries for simple signal quantification. Proof of concept experiments demonstrate both quantitative and multiplex assays in FEP MCF devices using a standard direct ELISA procedure and read using a flatbed scanner. This new multiplex immunoassay platform should find applications ranging from lab detection to point-of-care and field diagnostics

    Landscape of Epidermal Growth Factor Receptor Heterodimers in Brain Metastases

    HER2+ breast cancer patients have an elevated risk of developing brain metastases (BM), despite adjuvant HER2-targeted therapy. The mechanisms underpinning this reduced intracranial efficacy are unclear. We optimised the in situ proximity ligation assay (PLA) for detection of the high-affinity neuregulin-1 receptor, HER2-HER3 (a key target of pertuzumab), in archival tissue samples and developed a pipeline for high throughput extraction of PLA data from fluorescent microscope image files. Applying this to a large BM sample cohort (n = 159) showed that BM from breast, ovarian, lung and kidney cancers have higher HER2-HER3 levels than other primary tumour types (melanoma, colorectal and prostate cancers). HER2 status, and tumour cell membrane expression of pHER2(Y1221/1222) and pHER3(Y1222) were positively, but not exclusively, associated with HER2-HER3 frequency. In an independent cohort (n = 78), BM had significantly higher HER2-HER3 levels than matching primary tumours (p = 0.0002). For patients who had two craniotomy procedures, HER2-HER3 dimer levels were lower in the consecutive lesion (n = 7; p = 0.006). We also investigated the effects of trastuzumab and pertuzumab on five different heterodimers in vitro: HER2-EGFR, HER2-HER4, HER2-HER3, HER3-HER4, HER3-EGFR. Treatment significantly altered the absolute frequencies of individual complexes in SKBr3 and/or MDA-MB-361 cells, but in the presence of neuregulin-1, the overall distribution was not markedly altered, with HER2-HER3 and HER2-HER4 remaining predominant. Together, these findings suggest that markers of HER2 and HER3 expression are not always indicative of dimerization, and that pertuzumab may be less effective at reducing HER2-HER3 dimerization in the context of excess neuregulin

    Automatic activation, interference and facilitation effects in persons with aphasia and normal adult controls on experimental CRTT-R-Stroop tasks

    The current study investigated the effects of several color word congruent and incongruent “Stroop” tasks, within the context of a reading comprehension test (CRTT-R-wf-Stroop), in persons with aphasia (PWA) (N=25) and normal adults (NA) (N=29). Reading times, percentage of correct responses and CRTT-R-wf scores were examined for the color words. Both groups demonstrated significant vigilance and interference effects on RT ratios reflecting costs in sustained attention, interference/suppression effects and attentional switching. Both groups showed a facilitation effect on the CRTT-R-wf score. Unlike the NA, the PWA showed no attentional effects for the number of correct response on the color adjectives

    A crossover randomised controlled trial of oral mandibular advancement devices for obstructive sleep apnoea-hypopnoea (TOMADO)

    Rationale Mandibular advancement devices (MADs) are used to treat obstructive sleep apnoea-hypopnoea syndrome (OSAHS) but evidence is lacking regarding their clinical and cost-effectiveness in less severe disease. Objectives To compare clinical- and cost-effectiveness of a range of MADs against no treatment in mild to moderate OSAHS. Measurements and methods This open-label, randomised, controlled, crossover trial was undertaken at a UK sleep centre. Adults with Apnoea-Hypopnoea Index (AHI) 5–<30/h and Epworth Sleepiness Scale (ESS) score ≥9 underwent 6 weeks of treatment with three nonadjustable MADs: self-moulded (SleepPro 1; SP1); semi-bespoke (SleepPro 2; SP2); fully-bespoke MAD (bMAD); and 4 weeks no treatment. Primary outcome was AHI scored by a polysomnographer blinded to treatment. Secondary outcomes included ESS, quality of life, resource use and cost. Main results 90 patients were randomised and 83 were analysed. All devices reduced AHI compared with no treatment by 26% (95% CI 11% to 38%, p=0.001) for SP1, 33% (95% CI 24% to 41%) for SP2 and 36% (95% CI 24% to 45%, p<0.001) for bMAD. ESS was 1.51 (95% CI 0.73 to 2.29, p<0.001, SP1) to 2.37 (95% CI 1.53 to 3.22, p<0.001, bMAD) lower than no treatment (p<0.001 for all). Compliance was lower for SP1, which was the least preferred treatment at trial exit. All devices were cost-effective compared with no treatment at a £20 000/quality-adjusted life year (QALY) threshold. SP2 was the most cost-effective up to £39 800/QALY. Conclusions Non-adjustable MADs achieve clinically important improvements in mild to moderate OSAHS and are cost-effective
